چکیده

To investigate the radiation response of hepatoma, SMMC-7721 cells were irradiated with 60 Co γ-rays. Initial chromatid breaks were measured by counting the number of chromatid breaks and isochromatid breaks. A dose-dependent increase in radiation-induced chromatid/isochromatid breaks was observed in G1 and G2 phase respectively. A good relationship was found between cell survival and chromatin breaks. This study implied that low LET radiation-induced chromatid/isochromatid breaks can be possibly used as a good predictor of radio sensitivity of SMMC-7721 hepatoma cells.

Biological Dose Assessment by Chemical Induction of Premature Chromosome Condensation

Dose assessment of radiation victims is a key element in medical management of radiation accidents. At high radiation doses (> 6 Gy), dicentric assay lose its efficiency and premature chromosome condensation (PCC) assay was proposed to overcome the restrictions of dicentric assay. It was shown that PCC ring is more suitable and simpler than dicentric for biodosimetry in high radiation doses. In...

Construction of a dose – response curve by induction of premature chromosome condensation for biological dosimetry

Premature Chromosome Condensation (PCC) appears to have a possible utility for biological dosimetry purposes. The PCC technique may be adapted for cases of suspicion of overexposure where sampling is performed at least one day after an accident. For this purpose, human blood samples were exposed in vitro to 60Co up to 10 Gy and the PCC technique was performed immediately after irradiation. Anal...

Induction of premature chromosome condensation (PCC) by calyculin A for biodosimetry

Background: Premature chromosome condensation (PCC) is a method for interphase chromosome analysis in biodosimetry. This study was performed to verify the usefulness of PCC induced by calyculin A in human peripheral blood lymphocytes (pbl) for biological dosimetry and possible construction of dose-response curve. Materials and Methods: Peripheral blood was obtained from a healthy donor and expo...

ارزیابی آسیب‌های کروموزومی بیماران مبتلا به سرطان مری پیش و پس از رادیوتراپی با استفاده از روش آزمون میکرونوکلئی

Background: Radiotherapy can cause DNA damage in normal cells, misrepaired or unrepaired double strand breaks in DNA lead to chromosomal breaks. As a result patient experience early and late effects in normal tissue during and after radiotherapy. Cytogenetic techniques can be used as a cancer predictive assay because there is an association between chromosome abnormalities and the risk of devel...

Bleomycin-induced damage in prematurely condensed chromosomes and its relationship to cell cycle progression in CHO cells.

between the cytotoxic effects of these agents and the incidence of chromosome aberrations in the treated cells (2,4, 15). However, BLM treatment severely inhibits the progression of cells into mitosis. The probability that the extensively damaged cells might not be reaching mitosis makes it difficult to assay the chromosome damage accurately by the conven tional techniques. Recently, we have sh...
